IDENTIFICATION OF DIFFERENTIALLY EXPRESSED MIRNAS USING RNA-SEQ DATA: A CASE STUDY IN MALE AND FEMALE CHILDREN WITH MEDULLOBLASTOMA / Nihal HABIB
Titre : IDENTIFICATION OF DIFFERENTIALLY EXPRESSED MIRNAS USING RNA-SEQ DATA: A CASE STUDY IN MALE AND FEMALE CHILDREN WITH MEDULLOBLASTOMA Type de document : thèse Auteurs : Nihal HABIB, Auteur Année de publication : 2020 Langues : Anglais (eng) Mots-clés : miRNA transcriptomics Gene Expression regulation Differential expression Medulloblastoma Cancer RNA-seq Bioinformatics RSubread DESeq2 miARN transcriptomique régulation de l'expression génique expression différentielle médulloblastome cancer RNA-seq bioinformatique RSubread DESeq2. Résumé : microRNAs (miRNAs) are small non-coding RNA molecules that contain approximately 22 nucleotides and they play important roles in regulating gene expression in animals, plants and some viruses. Their regulation has been implicated in the pathogenesis of a number of human pathologies including cancers. In the present work, we aimed to investigate expression profiles of micro-RNA (miRNA) in patients with Medulloblastoma cancer, which is the most common type of primary brain cancer in children. It starts in the lower back part of the brain, called the cerebellum which is involved in muscle coordination, balance and movement. . The number of methods and softwares for differential expression analysis from RNA-Seq data increased rapidly. In this comparative study, we identified and analysed Human sex-biased Micro-RNAs in infants by examining a transcriptome dataset of patients with Medulloblastoma. This was done using RSubread as a tool for mapping and its command “featurescounts” to quantify the miRNAs, and DESeq2 bioinformatics package to conduct a differential analysis of miRNAs between infant males and females. Consequently, 147 miRNAs were found differentially expressed with different abundances between the sexes. The results further highlight the applicability of miRNA expression patterns as potential markers of human Medulloblastoma cancer and most probably other cancers in children.
